    Area of Science:

    • Prenatal diagnosis
    • Cytogenetics
    • Human genetics

    Background:

    • Chorionic villi sampling (CVS) is a prenatal diagnostic technique.
    • Obtaining direct chromosome preparations from CVS is challenging.
    • Accurate genetic information is crucial for reproductive decision-making.

    Purpose of the Study:

    • To evaluate the efficacy of direct chromosome preparations from chorionic villi.
    • To optimize hypotonic treatment and cell dispersal for CVS samples.
    • To assess the quality of G-banded chromosomes obtained from CVS.

    Main Methods:

    • Chorionic villi sampling performed on 52 patients undergoing elective pregnancy termination.
    • Direct chromosome preparations attempted on 42 successful villi samples.

    Related Experiment Videos

  • Hypotonic treatment using a 2:1 mixture of 0.075 M potassium chloride and 1% sodium citrate.
  • Cell dispersal achieved with 40% acetic acid.
  • Main Results:

    • Direct chromosome preparations were successful in 41 out of 42 villi samples.
    • Chromosomes exhibited good morphology.
    • G-banding analysis was successfully performed, yielding clear G-bands.

    Conclusions:

    • Direct chromosome preparations from chorionic villi are a viable method for genetic analysis.
    • The optimized hypotonic treatment and cell dispersal protocols enhance chromosome quality.
    • This technique provides reliable genetic information for prenatal diagnosis.
