Signal transducer and activator of transcription 5a (Stat5a) promotes mouse type X collagen gene (Col10a1) expression and chondrocyte hypertrophy. Stat5a interacts with the Col10a1 cis-enhancer, regulating Col10a1 expression and hypertrophic differentiation.
Area of Science:
- Molecular Biology
- Cell Biology
- Developmental Biology
Background:
- Transcription factors (TFs) regulate hypertrophic chondrocyte-specific gene expression.
- Mouse type X collagen gene (Col10a1) expression is crucial for chondrocyte hypertrophy.
- Previous studies identified TFs interacting with Col10a1 promoters.
Purpose of the Study:
- Investigate the role of signal transducer and activator of transcription 5a (Stat5a) as a binding factor for the Col10a1 cis-enhancer.
- Elucidate the mechanism by which Stat5a controls Col10a1 gene expression.
- Determine Stat5a's influence on chondrocyte hypertrophic differentiation.
Main Methods:
- Transcription factor affinity prediction (TRAP) analysis identified Stat5a as a potential regulator.
- Quantitative reverse transcription polymerase chain reaction (qRT-PCR), western blot, and immunohistochemistry (IHC) verified Stat5a expression.
- Cellular experiments involved Stat5a knockdown/overexpression and dual-luciferase reporter assays.
- Chondrocyte differentiation was assessed via staining and marker gene expression analysis.
Main Results:
- Stat5a and Col10a1 expression are positively correlated in hypertrophic chondrocytes.
- Stat5a knockdown decreased Col10a1 expression; Stat5a overexpression enhanced it.
- Stat5a potentiated reporter activity driven by the Col10a1 promoter/enhancer.
- Stat5a increased alkaline phosphatase activity and expression of hypertrophic markers like Runx2.
Conclusions:
- Stat5a acts as a positive regulator of Col10a1 expression.
- Stat5a promotes chondrocyte hypertrophic differentiation.
- Stat5a likely interacts with the Col10a1 cis-enhancer to mediate its effects.
