The Tumor Microenvironment02:17

The Tumor Microenvironment

6.7K
Every normal cell or tissue is embedded in a complex local environment called stroma, consisting of different cell types, a basal membrane, and blood vessels. As normal cells mutate and develop into cancer cells, their local environment also changes to allow cancer progression. The tumor microenvironment (TME) consists of a complex cellular matrix of stromal cells and the developing tumor. The cross-talk between cancer cells and surrounding stromal cells is critical to disrupt normal tissue...

Tumor Immunotherapy01:27

Tumor Immunotherapy

567
Immunotherapy is a treatment that boosts or manipulates the immune system to fight diseases, including cancer. For instance, by stimulating an immune response through vaccinations against viruses that cause cancers, like hepatitis B virus and human papillomavirus, these diseases can be prevented. Nonetheless, some cancer cells can avoid the immune system due to their rapid mutation and division. The immune response to many cancers involves three phases: elimination, equilibrium, and escape.

Area of Science:

  • Cancer immunology
  • Computational biology
  • Bioinformatics

Background:

  • Tumor cells and immune cells communicate, influencing tumor immunity.
  • Existing knowledge on tumor-immune cell (TIC) communications is fragmented across numerous studies.
  • Systematic characterization of TIC communications across various cancers is challenging.

Purpose of the Study:

  • To develop a comprehensive database, TICCom, for modeling TIC communications.
  • To consolidate experimentally validated and computationally predicted TIC interactions.
  • To provide a centralized resource for studying TIC communications in cancer.

Main Methods:

  • Collected 739 experimentally-validated or manually-curated TIC interactions from over 3,000 literature sources.
  • Inferred 4,537,709 predicted interactions using six computational algorithms.
  • Reanalyzed 32 scRNA-seq and bulk RNA-seq datasets across 25 cancer types.
  • Integrated ligand-receptor interactions and functional information.

Main Results:

  • TICCom models communications between tumor cells and 14 types of immune cells.
  • The database contains 14,190 human and 3,650 mouse integrated ligand-receptor interactions.
  • Includes supplementary functional information for these interactions.

Conclusions:

  • TICCom serves as a valuable, comprehensive resource for investigating tumor-immune cell communications.
  • Facilitates systematic characterization of TIC interactions across diverse cancer types.
  • Aids researchers in understanding the complex interplay shaping tumor immunity.
