    Summary

    The enzyme inhibition method is preferred for quantifying urinary D-glucaric acid over the glyoxylic acid method. It is more reliable due to less interference and is less labor-intensive.

    Background:

    • Urinary D-glucaric acid is a biomarker for certain metabolic processes.
    • Accurate quantification of D-glucaric acid is essential for clinical and research applications.
    • Existing spectrophotometric methods have limitations in accuracy and labor intensity.

    Purpose of the Study:

    • To evaluate and compare the relative usefulness of two spectrophotometric methods for quantifying urinary D-glucaric acid.
    • To identify the most reliable and efficient method for routine analysis.

    Main Methods:

    • Enzyme inhibition method: Measures beta-D-glucuronidase inhibition by 1,4-glucarolactone derived from urinary D-glucaric acid.
    • Glyoxylic acid method: Involves ion exchange chromatography separation followed by oxidation of D-glucaric acid to glyoxylic acid.
    • Both methods were applied to urine samples from 61 healthy adults and standard solutions.

    Main Results:

    • Both methods yielded similar results for standard solutions with approximately 105% recovery.
    • Urinary D-glucaric acid concentrations were 3-10 times higher with the glyoxylic acid method compared to the enzyme inhibition method.
    • Higher values in the glyoxylic acid method were attributed to interference from ascorbic acid and other compounds.

    Conclusions:

    • The modified enzyme inhibition method is preferable for quantifying urinary D-glucaric acid.
    • The enzyme inhibition method demonstrates greater specificity and is less labor-intensive than the glyoxylic acid method.
    • Interference from ascorbic acid significantly impacts the accuracy of the glyoxylic acid method.
