    Summary

Thyroid hormone thyroglobulin (Tg) release is significantly elevated in Graves' disease patients, while its metabolic clearance rate remains normal. This study investigated Tg kinetics in healthy individuals and those with Graves' disease.

    Area of Science:

    • Endocrinology
    • Thyroidology
    • Pharmacokinetics

    Background:

    • Thyroglobulin (Tg) is detectable in normal circulation.
    • Elevated serum Tg is observed in thyroid disorders like Graves' disease.
    • Tg metabolism in Graves' disease is not well understood.

    Purpose of the Study:

    • To investigate the kinetics of human thyroglobulin (Tg) in normal men and hyperthyroid patients with Graves' disease.
    • To compare Tg metabolism and release rates between healthy subjects and those with Graves' disease.

    Main Methods:

    • A kinetic study involving the intravenous injection of 10 mg of Tg as a bolus dose.
    • Blood sample collection at various intervals up to 72 hours post-injection.
    • Serum Tg concentrations measured using RIA; kinetic parameters calculated via single compartmental analysis.

    Related Experiment Videos

    Main Results:

    • Normal subjects: Mean Tg half-life 29.6 ± 2.8 h, MCR 268.9 ± 87.8 ml/h/60 kg, release rate 100.3 ± 50.2 µg/day/60 kg.
    • Graves' disease patients: Increased Tg release rates in all six patients.
    • Tg half-life and MCR were generally within normal ranges, even in hyperthyroid patients.

    Conclusions:

    • Thyroglobulin release is significantly increased in hyperthyroid Graves' disease patients.
    • Thyroglobulin metabolism, as indicated by half-life and MCR, appears similar in Graves' disease patients and normal subjects.
    • The study elucidates altered Tg dynamics in Graves' disease, highlighting increased secretion rather than impaired clearance.
