Single Nucleotide Polymorphisms-SNPs01:05

Single Nucleotide Polymorphisms-SNPs

15.3K
A single nucleotide polymorphism or SNP is a single nucleotide variation at a specific genomic position in a large population. It is the most prevalent type of sequence variation found in the human genome. Point mutations that occur in more than 1% of the population qualify as SNPs. These are present once every 1000 nucleotides on an average in the human genome. Replacement of a purine with another purine (A/G) or a pyrimidine with another pyrimidine (C/T) is known as a transition. In contrast,...

Area of Science:

  • Oral health and infectious diseases
  • Genetics and epidemiology

Background:

  • Observational studies suggest a link between periodontitis and Coronavirus disease (COVID-19).
  • The causal relationship between periodontitis and COVID-19 remains unclear.

Purpose of the Study:

  • To determine the causal influence of COVID-19 on periodontitis using bidirectional Mendelian randomization (MR).
  • To investigate the potential causal effect of periodontitis on COVID-19 outcomes.

Main Methods:

  • Utilized large-scale genome-wide association study data for COVID-19 and periodontitis.
  • Employed inverse variance weighting, MR-Egger, weighted median, and MR-PRESSO for causal effect estimation.
  • Conducted sensitivity analyses including Cochran's Q test and MR-PRESSO to assess pleiotropy.

Main Results:

  • No causal relationship was found between periodontitis and COVID-19 hospitalization, susceptibility, or disease severity.
  • Bidirectional MR analysis indicated no significant causal effect in either direction.
  • Sensitivity analyses confirmed the robustness of the findings, suggesting minimal impact from horizontal pleiotropy.

Conclusions:

  • Periodontitis is not causally linked to an increased risk of COVID-19 hospitalization, susceptibility, or severity.
  • Further investigation is warranted to explore potential underlying biological mechanisms or shared risk factors.
