    Summary

    Even brief exposure to ethanol during critical brain development periods can cause lasting damage. This study shows that episodic alcohol exposure can be as harmful as chronic exposure for developing brains.

    Background:

    • The developing brain's susceptibility to ethanol is not fully understood.
    • Previous research has not clearly defined the critical windows for ethanol-induced brain damage.

    Purpose of the Study:

    • To investigate if low blood ethanol concentration (BEC) or brief ethanol exposure during the brain growth spurt causes permanent brain damage.
    • To compare the effects of chronic versus episodic ethanol exposure on brain development.

    Main Methods:

    • Rats were administered two doses of ethanol: 4 g/kg daily from postnatal days 6-16, or 6 g/kg on postnatal day 6 only.
    • Evaluated whole brain weight, cerebellar weight, motor coordination (balancing ability), and cerebellar cell counts at postnatal days 17 and 70.

    Main Results:

    • Both ethanol exposure groups showed significant reductions in whole brain weight and cerebellar weight compared to controls.
    • Ethanol exposure led to impaired balancing ability and decreased cerebellar cell numbers.
    • Episodic exposure on day 6 resulted in more severe cerebellar weight reduction than chronic exposure during days 6-16.

    Conclusions:

    • Both chronic and episodic ethanol exposure during the brain growth spurt can cause permanent brain damage.
    • Episodic ethanol exposure during peak brain development may be as detrimental as continuous exposure.
    • These findings highlight the vulnerability of the developing brain to alcohol, even at lower concentrations or shorter durations.
