DNA barcoding using internal transcribed spacers (ITS) offers accurate identification for Korean Caryophyllaceae plants. While ITS showed higher accuracy, combining it with chloroplast regions improved resolution, highlighting the need for taxonomic review in Korea.
Area of Science:
- Botany
- Molecular Biology
- Biodiversity Studies
Background:
- The Caryophyllaceae family is significant in Korea, encompassing ornamental, medicinal, and endangered species.
- Accurate identification of plant species is crucial for conservation, resource management, and scientific research.
Purpose of the Study:
- To evaluate the efficacy of DNA barcoding for identifying Korean Caryophyllaceae species.
- To compare the discriminatory power of different DNA markers for this plant family.
Main Methods:
- Sequencing of three chloroplast DNA regions (rbcL, matK, psbA-trnH) and nuclear ribosomal internal transcribed spacers (ITS) from 78 taxa (n=215).
- Phylogenetic analysis using neighbor-joining trees to assess identification accuracy and resolution.
- Comparison of identification success rates between chloroplast markers and ITS, and their combined use.
Main Results:
- Internal transcribed spacers (ITS) provided higher identification accuracy (>73%) compared to chloroplast regions (<62%).
- Combining rbcL and ITS yielded the highest resolution (77.6%), though accuracy varied by genus.
- Five genera (Eremogone, Minuartia, Pseudostellaria, Sagina, Stellaria) were successfully identified, while five others (Cerastium, Gypsophila, Dianthus, Silene, Spergularia) showed low resolution.
Conclusions:
- DNA barcoding, particularly using ITS, is effective for identifying Korean Caryophyllaceae.
- Low resolution in certain genera suggests potential taxonomic complexities or issues with marker performance.
- The findings underscore the necessity for taxonomic reconsideration of Caryophyllaceae species in Korea.
