Area of Science:

  • Biotechnology
  • Immunology
  • Drug Discovery

Background:

  • Antibodies are a significant and expanding class of therapeutic drugs.
  • Phage display technology is a key method for antibody discovery.
  • Current phage display methods often yield complex antibody pools where abundance doesn't correlate with function.

Purpose of the Study:

  • To address the limitations of standard phage display enrichment.
  • To introduce deep mining technologies for uncovering low-abundant, functionally important antibody clones.
  • To enable comprehensive analysis of antibody pools generated by phage display.

Main Methods:

  • Description of two deep mining techniques: protein depletion and antibody blocking.
  • Application of these methods to antibody pools derived from phage display selection.
  • Adaptability of methods for both single antigen targets and complex mixtures like cells and tissues.

Main Results:

  • Deep mining enables the discovery of antibodies missed by standard screening.
  • Protein depletion and antibody blocking can reveal functionally significant low-abundant clones.
  • These methods enhance the utility of phage display output.

Conclusions:

  • Standard phage display screening may overlook crucial antibody candidates.
  • Deep mining strategies are essential for maximizing the potential of phage display.
  • Protein depletion and antibody blocking offer robust approaches for comprehensive antibody pool analysis.
