    Background:

    • Radiation therapy can cause lung damage.
    • Actinomycin D is a drug with potential radioprotective properties.
    • Previous research on its effects has yielded conflicting results.

    Purpose of the Study:

    • To evaluate the effect of actinomycin D on radiation-induced lung damage in mice.
    • To determine if the timing of actinomycin D administration influences its efficacy.

    Main Methods:

    • CBA mice received single thoracic X-ray doses.
    • Actinomycin D (0.4 mg/kg) was administered at different time points: 4 weeks before, immediately after, or 16 weeks after irradiation.
    • Respiration rate was measured using whole-body plethysmography to assess lung damage.
    • Dose-response curves were generated from 12 to 40 weeks post-irradiation.

    Main Results:

    • Actinomycin D administration did not significantly alter respiration rates in mice subjected to thoracic irradiation.
    • No significant protective or detrimental effect of actinomycin D on radiation-induced lung damage was observed in this study.
    • Experimental findings were compared with existing literature, highlighting discrepancies.

    Conclusions:

    • Actinomycin D does not appear to mitigate radiation-induced lung damage in mice under the conditions tested.
    • Further research is needed to reconcile conflicting results and clarify the role of actinomycin D in radiation injury.
    • The timing of drug administration did not influence the observed lack of effect.
