Inking cell blocks improves automatic detection (ADS) in digital pathology, particularly for immunostained slides. This enhances diagnostic accuracy by ensuring complete whole slide image (WSI) scanning, reducing false negatives.

Background:

  • Incomplete whole slide images (WSIs) due to poor automatic slide detection (ADS) can lead to missed diagnoses.
  • Cell blocks present challenges for automated scanning, potentially resulting in false negatives.

Purpose of the Study:

  • To evaluate if inking cell blocks enhances ADS for improved WSI generation.
  • To assess the impact of ink color on scanning efficiency and image quality.

Main Methods:

  • Two tests were conducted on 15 cell blocks each.
  • Test 1: Compared black-inked (1HB) vs. green-inked (1HG) halves for H&E staining.
  • Test 2: Compared black-inked (2HB) vs. unstained (2HN) halves for HE, PAS, and CKAE1AE3 immunostaining.

Main Results:

  • Black ink interfered less with observation than green ink (2.2% vs. 44.4%).
  • For H&E and PAS stains, both inked and unstained halves achieved total ADS with similar scanning times and file sizes.
  • For CKAE1AE3 immunostaining, black-inked cell blocks significantly increased ADS (93.3% vs. 46.7%), scanning time, and file size compared to unstained halves.

Conclusions:

  • Inking cell blocks, especially with black ink, significantly improves automatic slide detection for immunostained samples.
  • While increasing scanning time and file size, inking enhances WSI completeness and diagnostic safety in digital pathology workflows.
  • This method offers a practical solution to mitigate false negatives arising from incomplete slide scanning.
