Summary

This study maps human mesenchymal stem cell differentiation to understand osteosarcoma (OS) heterogeneity. A stem-like cell signature in OS tumors correlates with poorer survival, offering therapeutic targets.

Area of Science:

  • Oncology
  • Stem Cell Biology
  • Genetics

Background:

  • Osteosarcoma (OS) exhibits genetic and intratumoral heterogeneity, complicating drug development and the study of cell fate.
  • Understanding cell differentiation, plasticity, and fate is crucial for OS tumor grade, metastasis, and survival.

Conclusions:

  • This study provides the first accurate quantification of osteosarcoma differentiation and lineage commitment.
  • A stem-like transcriptomic signature is associated with adverse survival in osteosarcoma.
  • Understanding differentiation pathways is a critical prerequisite for developing targeted osteosarcoma therapies.
