    Summary

pH-sensitive liposomes show potential for lung delivery of lipophilic drugs, accumulating significantly in lung tissue. pH-insensitive liposomes distribute mainly to the liver and spleen.

    Area of Science:

    • Nanotechnology
    • Pharmacology
    • Biomedical Engineering

    Background:

    • Liposomes are versatile drug delivery systems.
    • Understanding liposome tissue distribution is crucial for optimizing drug delivery.
    • pH-sensitive liposomes offer potential for targeted drug release.

    Purpose of the Study:

    • To investigate the tissue distribution of pH-sensitive and pH-insensitive liposomes.
    • To evaluate the impact of liposome composition and surface modification on biodistribution.
    • To determine the potential of pH-sensitive liposomes for targeted drug delivery, particularly to the lungs.

    Main Methods:

    • Preparation of pH-sensitive (dioleoylphosphatidylethanolamine/oleic acid) and pH-insensitive (dioleoylphosphatidylcholine/oleic acid) liposomes.
    • Injection of liposomes into C3H and Balb/c mice.
    • Monitoring lipid and aqueous content distribution using radiolabeled markers ([3H]cholestanyl ether and 125I-tyraminyl-inulin).
    • Assessment of liposome behavior with and without surface-bound monoclonal antibodies.

    Main Results:

    • pH-insensitive liposomes were rapidly cleared, accumulating primarily in the liver and spleen.
    • Surface-modified pH-insensitive liposomes showed similar distribution patterns.
    • pH-sensitive liposomes exhibited leakage in mouse plasma.
    • The lipid component of pH-sensitive liposomes showed significant accumulation (50-70%) in the lungs for up to 6 hours.
    • Monoclonal antibodies did not alter the distribution profile of pH-sensitive liposomes.

    Conclusions:

    • pH-sensitive liposomes are not effective for aqueous drug delivery due to leakage.
    • The lung accumulation of pH-sensitive liposomes suggests their potential for delivering lipophilic drugs to lung tissue.
    • Liposome composition significantly influences tissue distribution and potential therapeutic applications.
