Dengue virus (DENV) infection increases the stress modulator Angiogenin in host cells. Inhibiting Angiogenin with Ivermectin reduces DENV replication, suggesting its role in viral pathogenesis.
Area of Science:
- Virology
- Molecular Biology
- Cellular Stress Response
Background:
- Dengue virus (DENV) infection is a global health threat.
- DENV pathogenesis heavily relies on host-cellular machinery.
- Mechanistic details of DENV-host interactions remain incompletely characterized.
Purpose of the Study:
- To characterize the mechanistic basis of DENV-induced stress on host cells.
- To investigate the role of the stress modulator ribonuclease Angiogenin in DENV infection.
Main Methods:
- Quantification of Angiogenin levels in DENV-infected cells.
- siRNA-mediated knockdown of Angiogenin in DENV-infected and mock-infected cells.
- Treatment of Huh7 cells with Ivermectin prior to DENV infection.
Main Results:
- Angiogenin levels were upregulated in DENV-infected cells, increasing with viral replication.
- siRNA knockdown of Angiogenin was unsuccessful in DENV-infected cells.
- Ivermectin treatment significantly reduced DENV replication and Angiogenin levels in later stages.
Conclusions:
- Angiogenin upregulation is linked to DENV replication.
- Angiogenin modulation plays a crucial role in DENV replication and pathogenesis.
- Targeting Angiogenin may offer a therapeutic strategy against DENV.
