Formation of Lipopolysaccharides

Lipopolysaccharides (LPS) are crucial components of the outer membrane of Gram-negative bacteria, serving both structural and functional roles. It contributes to membrane stability and protects bacteria from host immune responses. LPS is composed of three major regions—lipid A, a core oligosaccharide, and an O antigen. The biosynthesis and assembly of LPS involve a highly coordinated set of enzymatic reactions and transport mechanisms. Additionally, LPS is recognized as an endotoxin,...

Bacterial Phylum Actinobacteria

Coryneform bacteria are gram-positive, aerobic, nonmotile rods that exhibit irregular, club-shaped, or V-shaped arrangements. Their V-shape results from snapping division, where the inner cell wall layer forms the cross-wall, while the outer layer remains intact until it ruptures on one side, causing the daughter cells to bend away. The primary genera are Corynebacterium and Arthrobacter. Corynebacterium includes diverse species, ranging from saprophytes to pathogens like Corynebacterium...

Area of Science:

  • Phytochemistry
  • Natural Products Chemistry
  • Microbiology

Background:

  • Ganoderma species are a rich source of bioactive triterpenoids.
  • Triterpenoids from Ganoderma have demonstrated various pharmacological properties, including antimicrobial effects.
  • Investigating the chemical constituents of Ganoderma tsugae can lead to the discovery of novel compounds.

Purpose of the Study:

  • To isolate and characterize novel and known lanostane triterpenoids from the fruiting bodies of Ganoderma tsugae.
  • To evaluate the in vitro antibacterial activities of the isolated triterpenoids against pathogenic microorganisms.

Main Methods:

  • Phytochemical investigation of the 80% ethanol extract of Ganoderma tsugae fruiting bodies.
  • Isolation and structural elucidation of compounds using 1D and 2D NMR spectroscopy (1H-1H COSY, HMQC, HMBC).
  • In vitro antibacterial activity testing against six pathogenic microorganisms using Minimum Inhibitory Concentration (MIC) values.

Main Results:

  • Two new lanostane triterpenoids, 7,11-dioxo-3β-acetyloxy-26,27-dihydroxy-lanosta-8,24-diene (1) and 7,20-dioxo-3β-acetyloxy-11β,15α-dihydroxy-22,23,24,25,26,27-hexanorlanosta-8-ene (2), were isolated.
  • One known lanostane triterpenoid, ganodermanontriol (3), was also identified.
  • Compound 3 exhibited antibacterial activity against three Gram-positive bacteria with MIC values below 30 μg/mL.

Conclusions:

  • The study successfully identified novel and known lanostane triterpenoids from Ganoderma tsugae.
  • Ganodermanontriol (3) demonstrates potential as an antibacterial agent against specific Gram-positive pathogens.
  • Further research into the structure-activity relationship of these triterpenoids could yield new antimicrobial drug leads.
