Bone marrow mesenchymal stem cell-derived exosomes (BMSC-Exo) alleviate diabetic kidney disease by reducing apoptosis and inflammation. This study shows BMSC-Exo treatment improves kidney structure and function in diabetic rats.
Area of Science:
- Regenerative Medicine
- Cell Biology
- Nephrology
Background:
- Diabetic kidney disease (DKD) is a major complication of diabetes.
- Bone marrow mesenchymal stem cell-derived exosomes (BMSC-Exo) have shown anti-inflammatory effects.
- The therapeutic potential of BMSC-Exo for DKD requires further investigation.
Purpose of the Study:
- To investigate the therapeutic effect of BMSC-Exo on DKD in a rat model.
- To elucidate the underlying mechanisms of BMSC-Exo in treating DKD.
Main Methods:
- A DKD rat model was established using streptozotocin and a high-fat diet.
- BMSC-Exo were administered weekly via tail vein injection for 12 weeks.
- Kidney pathology, apoptosis, inflammation markers (TNF-α, NF-κB), and biochemical parameters (GLU, Cr, BUN) were assessed.
Main Results:
- BMSC-Exo treatment significantly alleviated renal structural damage and reduced apoptotic cells in diabetic rats.
- BMSC-Exo modulated apoptosis-related proteins (Bax, Cleaved Caspase 9, Cleaved caspase 3).
- BMSC-Exo treatment decreased TNF-α and NF-κB (p65) expression, and improved GLU, Cr, and BUN levels.
Conclusions:
- BMSC-Exo demonstrate a therapeutic effect on DKD.
- BMSC-Exo may exert their protective effects by inhibiting renal cell apoptosis and inflammation.
