Antibody Structure and Classes01:25

Antibody Structure and Classes

931
Antibodies, also known as immunoglobulins, are produced by B cells in response to foreign substances, such as bacteria and viruses. These proteins are critical for recognizing and neutralizing these substances, protecting the body from potential harm.
The basic structure of an antibody consists of four protein chains: two identical heavy chains and two identical light chains. These chains are held together by disulfide bonds and other non-covalent interactions, forming a Y-shaped structure.

Area of Science:

  • Biotechnology
  • Proteomics
  • Immunology

Background:

  • The YCharOS initiative systematically characterizes antibodies against the human proteome.
  • Utilizes techniques like Western blot, immunoprecipitation, and immunofluorescence for antibody validation.

Discussion:

  • Poorly performing antibodies significantly impact research reproducibility.
  • Open science collaboration with industry partners offers an innovative solution to antibody quality issues.
  • Data reveals withdrawn or altered antibodies, highlighting the need for better validation.

Key Insights:

  • YCharOS has characterized 812 antibodies and 78 proteins, making data publicly available.
  • High-performing antibodies have been identified for many proteins, but this is a small fraction of the market.
  • Researchers must adapt procurement and usage practices based on validated data.

Outlook:

  • YCharOS data is accessible via Zenodo, F1000 articles, PubMed, and the Antibody Registry.
  • The initiative aims to improve antibody selection and usage, enhancing research integrity.
  • Wider adoption of validated antibody data is crucial for advancing scientific discovery.
