The membrane attack complex of complement induces permeability changes via thresholds in individual cells.

    Summary

Complement attack creates cell permeability and lysis thresholds in polymorphonuclear leucocytes (PMN). Adenosine inhibits membrane attack complex removal, increasing PMN lysis.

    Area of Science:

    • Immunology
    • Cell Biology

    Background:

    • The complement system plays a crucial role in innate immunity.
    • The membrane attack complex (MAC) is a key effector of complement-mediated cell lysis.

    Purpose of the Study:

    • To establish the existence of permeability and lytic thresholds in individual rat polymorphonuclear leucocytes (PMN) attacked by MAC.
    • To investigate the dynamics of cell recovery and lysis following complement activation.

    Main Methods:

    • Flow cytometry was utilized to quantify propidium iodide fluorescence in rat PMN.
    • Cells were exposed to the membrane attack complex (MAC) following complement pathway activation.

    Main Results:

    • Approximately 30-40% of PMN became permeable to molecules after MAC attack.

    Related Experiment Videos

  • Three distinct PMN populations were identified based on recovery status relative to permeability and lytic thresholds.
  • Individual PMN exhibited variable timing for permeability (7.5 ± 2.5 min) and lysis (11.5 ± 1.0 min) thresholds.
  • Conclusions:

    • Individual PMN display distinct permeability and lysis thresholds following complement attack.
    • Adenosine inhibited MAC removal, leading to increased PMN permeability and lysis.
