Background:

  • Gestational weight gain (GWG) has significant health implications for both mothers and offspring.

  • Genetic risk scores (GRS) offer a method to aggregate the effects of multiple single nucleotide polymorphisms (SNPs) for association studies.

Purpose of the Study:

  • To investigate the association between a genetic risk score (GRS) and gestational weight gain (GWG).
  • To analyze the relationship between specific SNPs (rs9939609, rs6548238, rs17782313, rs10938397, rs10913469, rs1137101, rs7799039, rs5443) and GWG in a Polish cohort.

Main Methods:

  • GWG was calculated for 342 healthy Polish women aged 19-45.
  • Eight selected SNPs in genes including FTO, TMEM18, MC4R, GNPDA2, SEC16B, LEPR, LEP, and GNB3 were genotyped.
  • Two GRS were computed: GRS1 (sum of all risk alleles) and GRS2 (sum of risk alleles for FTO, LEPR, LEP, GNB3).

Main Results:

  • A positive association was found between GRS2 and GWG (β = 0.12, p = 0.029).
  • Specific SNPs showed differential associations: TMEM18 (p=0.006, OR=2.6) and GNB3 (p<0.001, OR=3.3) with increased GWG.
  • GNPDA2 (p<0.001, OR=2.7) was associated with adequate GWG, and LEPR (p=0.011, OR=3.1) with decreased GWG.

Conclusions:

  • GRS2, along with genetic variants in TMEM18, GNB3, GNPDA2, and LEPR, are significantly associated with GWG.
  • These findings highlight the polygenic nature of GWG and the potential utility of GRS in understanding its genetic underpinnings.
