Area of Science:

  • Microbiology
  • Bacteriology
  • Taxonomy

Background:

  • The genus Pelagibacterium encompasses diverse bacterial species with various ecological roles.
  • Accurate classification of newly isolated microorganisms is crucial for understanding microbial diversity and function.

Purpose of the Study:

  • To characterize and classify a novel bacterial strain, YIM 151497T, isolated from a soil sample.
  • To determine the phylogenetic and taxonomic position of strain YIM 151497T within the genus Pelagibacterium.

Main Methods:

  • Isolation and cultivation of bacteria from soil samples.
  • Polyphasic taxonomic analysis including phenotypic, chemotaxonomic, and genotypic characterization.
  • 16S rRNA gene sequencing and whole-genome sequencing for phylogenetic analysis.
  • Determination of Average Nucleotide Identity (ANI) and digital DNA-DNA hybridization (dDDH) values.

Main Results:

  • Strain YIM 151497T is a Gram-stain-negative, yellow, aerobic, motile, rod-shaped bacterium.
  • Phylogenetic analysis indicated close relatedness to existing Pelagibacterium species, with ANI and dDDH values supporting novelty.
  • Physiological and biochemical tests revealed specific growth ranges (4-40°C, pH 4.0-10.0, 0-7% NaCl) and metabolic capabilities.
  • Genomic analysis showed a DNA G+C content of 60.7 mol% and identified key fatty acids and polar lipids.

Conclusions:

  • Strain YIM 151497T represents a novel species within the genus Pelagibacterium, named Pelagibacterium flavum sp. nov.
  • The findings contribute to the understanding of bacterial diversity in soil ecosystems.
  • This study provides a comprehensive description of a new bacterial species, aiding future research in microbial ecology and evolution.
