Imaging Biological Samples with Optical Microscopy01:18

Imaging Biological Samples with Optical Microscopy

Optical microscopy uses optic principles to provide detailed images of samples. Antonie van Leeuwenhoek designed the first compound optical microscope in the 17th century to visualize blood cells, bacteria, and yeast cells. In 1830, Joseph Jackson Lister created an essentially modern light microscope. The 20th century saw the development of microscopes with enhanced magnification and resolution.
In optical microscopy, the specimen to be viewed is placed on a glass slide and clipped on the stage...

Area of Science:

  • Microscopy and Image Analysis
  • Computational Biology
  • Biotechnology

Background:

  • Deep biological sample analysis often requires multi-modal, multi-time point, or multi-staining imaging.
  • Image alignment (registration) is crucial for co-localization analysis, enabling the fusion of complementary signals to understand biological processes.
  • Existing image registration methods lack universal applicability and user-friendly solutions, particularly for non-expert users.

Purpose of the Study:

  • To introduce DS4H Image Alignment (DS4H-IA), an open-source ImageJ/Fiji plugin for aligning 2D microscopy images.
  • To provide an extremely easy-to-use solution for aligning multi-modality, immunohistochemistry (IHC), and immunofluorescence (IF) images.
  • To offer a comprehensive resource including source code, standalone applications, tutorials, documentation, and sample datasets.

Main Methods:

  • Development of DS4H Image Alignment (DS4H-IA) as an ImageJ/Fiji plugin.
  • Implementation of an intuitive user interface for simplified image alignment.
  • Revision and comparison of existing 2D microscopy image alignment solutions.

Main Results:

  • DS4H-IA enables the alignment of multi-modal, IHC, and IF 2D microscopy images.
  • The plugin is designed for ease of use, requiring no specialized computer skills.
  • Open-source availability of DS4H-IA with cross-platform applications (MAC, Linux, Windows) and extensive support materials.

Conclusions:

  • DS4H Image Alignment (DS4H-IA) addresses the need for accessible and effective multi-modal image registration in biological research.
  • The plugin democratizes advanced image analysis techniques, facilitating co-localization studies.
  • Publicly available resources enhance the adoption and utility of DS4H-IA in diverse research settings.
