RNA Editing02:23

RNA Editing

RNA editing is a post-transcriptional modification where a precursor mRNA (pre-mRNA) nucleotide sequence is changed by base insertion, deletion, or modification. The extent of RNA editing varies from a few hundred bases, in mitochondrial DNA of trypanosomes, to a just single base, in nuclear genes of mammals. Even a single base change in the pre-mRNA can convert a codon for one amino acid into the codon for another amino acid or a stop codon. This type of re-coding can significantly affect the...

Alzheimer Disease l: Introduction

Alzheimer disease is a chronic, progressive, and irreversible neurodegenerative disorder and the most common cause of dementia in older adults. It leads to gradual neuronal loss, causing cognitive decline, behavioral changes, and loss of functional independence.Risk Factors and EtiologyThe disease is multifactorial. Age is the strongest risk factor, with prevalence doubling every 5 years after age 65. Genetic factors include mutations in genes such as APP, PSEN1, and PSEN2, which are associated...

Summary

Formalin-fixed paraffin-embedded (FFPE) brain tissue is valuable for research. However, robust Apolipoprotein-E (APOE) genotyping using PCR-RFLP is only reliable for FFPE tissue stored up to three years.

Area of Science:

  • Neuroscience
  • Genetics
  • Biotechnology

Background:

  • Formalin-fixed paraffin-embedded (FFPE) tissue banks are crucial research resources, particularly when linked with clinical data and psychometric testing.
  • Apolipoprotein-E (APOE) genotyping is essential for characterizing these valuable FFPE brain tissue samples.
  • The suitability of older FFPE tissues for genetic analysis, such as APOE genotyping, is a significant concern.

Discussion:

  • This study investigated the impact of storage duration on DNA quality for APOE genotyping in FFPE brain tissue.
  • Polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP) was employed to assess DNA integrity.
  • The findings highlight potential limitations of long-term FFPE storage for genetic analyses.

Key Insights:

  • Robust APOE genotyping using PCR-RFLP was achievable for FFPE brain tissue stored for a maximum of three years.
  • Tissue stored for longer periods, up to 19 years, showed reduced suitability for reliable APOE genotyping.
  • Prolonged storage of FFPE tissue blocks may have detrimental effects on DNA quality, impacting downstream genetic analyses.

Outlook:

  • Further research is needed to explore alternative methods for genotyping older FFPE samples.
  • Optimizing fixation and embedding protocols could potentially improve DNA preservation in FFPE tissues.
  • Developing standardized protocols for assessing DNA integrity in FFPE samples is crucial for future research endeavors.
