New ethosome and transethosome formulations effectively deliver Nutlin-3 to protect skin from UV damage. Transethosomes demonstrated superior delivery and UV protection compared to ethosomes.
Area of Science:
- Dermatology and Nanotechnology
- Photoprotection and Drug Delivery
Background:
- UV radiation induces skin damage, aging, and dermatological diseases.
- Nutlin-3, a p53 activator, inhibits UV-induced apoptosis in keratinocytes.
- Novel delivery systems are needed to enhance Nutlin-3's skin protective effects.
Purpose of the Study:
- To design and evaluate ethosomes and transethosomes as delivery systems for Nutlin-3.
- To assess the efficacy of these nanocarriers in protecting skin against UV-induced damage.
- To compare the skin penetration and photoprotective capabilities of ethosomes versus transethosomes.
Main Methods:
- Ethosomes and transethosomes loaded with Nutlin-3 were prepared.
- Vesicle characteristics (size, morphology, entrapment) were analyzed.
- In vitro drug release kinetics and ex vivo skin permeation were studied.
- Protective effects against UV radiation were evaluated in ex vivo skin explants.
Main Results:
- Both ethosomes and transethosomes efficiently entrapped Nutlin-3 (0.3% w/w).
- Transethosomes (146 nm) exhibited smaller, unilamellar vesicles compared to ethosomes (224 nm).
- Transethosomes showed significantly enhanced Nutlin-3 diffusion (250-fold higher) and permeability (7.4-fold higher) versus ethosomes.
- Ex vivo studies confirmed superior Nutlin-3 delivery and UV protection by transethosomes, preventing metalloproteinase activation and proliferation.
Conclusions:
- Ethosomes and transethosomes are effective nanocarriers for topical Nutlin-3 delivery.
- Transethosomes offer enhanced skin penetration and superior photoprotective efficacy compared to ethosomes.
- This study highlights the potential of transethosomes for developing advanced topical treatments against UV-induced skin damage.

Nucleotide Excision Repair
Cells are regularly exposed to mutagens—factors in the environment that can damage DNA and generate mutations. UV radiation is one of the most common mutagens and is estimated to introduce a significant number of changes in DNA. These include bends or kinks in the structure, which can block DNA replication or transcription. If these errors are not fixed, the damage can cause mutations, which in turn can result in cancer or disease depending on which sequences are...
