We developed a rapid cellulose-based method to purify double-stranded RNA (dsRNA) from virus-infected plant and fungal samples. This technique enables efficient high-throughput isolation and analysis for virus characterization.
Area of Science:
- Molecular Biology
- Virology
- Biochemistry
Background:
- Double-stranded RNA (dsRNA) are key replication intermediates for all RNA viruses.
- Accurate dsRNA purification and analysis are crucial for viral diagnosis and characterization.
- Cellulose is a known material for dsRNA isolation, binding it selectively in 15% ethanol.
Purpose of the Study:
- To describe a rapid, reliable, and high-throughput method for purifying dsRNA.
- To utilize a home-made micro-spin cellulose column for dsRNA isolation from plant and fungal cell lysates.
- To facilitate routine dsRNA analysis in virology research.
Main Methods:
- Developed a micro-spin column using cellulose for dsRNA purification.
- Applied the method to cell lysates from virus-infected plant tissues.
- Optimized ethanol concentration for selective dsRNA binding to cellulose.
Main Results:
- Successfully isolated dsRNA using the home-made cellulose column.
- The method proved to be rapid, labor-saving, and reliable.
- Enabled high-throughput dsRNA isolation suitable for routine analysis.
Conclusions:
- The described method offers an efficient approach for dsRNA purification from plant and fungal samples.
- This technique supports routine high-throughput dsRNA analysis for virus diagnosis and characterization.
- Home-made cellulose columns provide a cost-effective solution for dsRNA isolation.
