Long-term depression, or LTD, is one of the ways by which synaptic plasticity—changes in the strength of chemical synapses—can occur in the brain. LTD is the process of synaptic weakening that occurs over time between pre and postsynaptic neuronal connections.   • Reduced intracortical myelination (ICM) is implicated in depression, but its role as a precursor or consequence is unclear.

Purpose of the Study:

  • To investigate whether reduced intracortical myelination (ICM) is an early neural marker of vulnerability for major depressive disorder (MDD).
  • To examine ICM in unaffected offspring of parents with and without a history of depression.

Main Methods:

  • Acquired T1-weighted and T2-weighted MRI scans from 56 unaffected offspring of parents with depression and 63 unaffected offspring of parents without depression (ages 9-16).
  • Calculated test-retest reliability to assess scan quality.
  • Computed myelin maps for 68 cortical regions and analyzed data using mixed-effects modeling.

Main Results:

  • No significant differences in global or regional intracortical myelination (ICM) were found between high and low familial risk youths.
  • The pediatric sample exhibited high reliability for ICM measurements (ICC=0.79).

Conclusions:

  • Reduced intracortical myelination (ICM) does not appear to be a precursor of major depressive disorder (MDD).
  • Further research is recommended to explore ICM in familial high-risk youths throughout development.
