Purpose of the Study:

  • To investigate the adipogenic potential of BPA analogues: Tetra Methyl Bisphenol F (TMBPF), Bisphenol F (BPF), Bisphenol AP (BPAP), and fluorine-9-bisphenol (BHPF).
  • To compare the effects of these analogues with BPA on fat cell differentiation.

Main Methods:

  • Murine 3T3-L1 cells were treated with BPA and its analogues (0.01–20 µM) during differentiation.
  • Assessed lipid accumulation and expression of adipogenic markers (mRNA and protein).

  • TMBPF, BPF, and BPA significantly increased lipid accumulation and adipogenic gene expression (Lpl, Fabp4, Plin) at 1–20 µM.
  • TMBPF demonstrated a stronger adipogenic effect than BPA and other analogues.
  • TMBPF's effect on Fabp4 expression suggests peroxisome proliferator-activated receptor (PPAR) γ activation.

Conclusions:

  • BPF mimics BPA's effects, while TMBPF is the most potent adipogenic analogue.
  • TMBPF acts as a PPARγ agonist, indicating it is an obesogenic chemical and potential metabolic disruptor.
