Abstract

Insights

The miR-146a gene rs2910164 polymorphism is linked to multiple sclerosis (MS) susceptibility in Iranians. The rs2431697 polymorphism showed no association with MS in this population.

Area of Science:

  • Genetics
  • Immunology
  • Neurology

Background:

  • Multiple sclerosis (MS) is an autoimmune inflammatory disease of the central nervous system.
  • MicroRNA-146a (miR-146a) is a non-coding RNA that regulates immune responses.

Purpose of the Study:

  • To investigate the association between miR-146a gene polymorphisms (rs2910164 and rs2431697) and MS risk in the Iranian population.

Main Methods:

  • Genotyping of rs2910164 and rs2431697 polymorphisms using PCR-RFLP and Tetra-primer ARMS-PCR.
  • Case-control study with 60 MS patients and 100 healthy controls.
  • Statistical analysis using the chi-squared test and Hardy-Weinberg equilibrium evaluation.

Main Results:

  • The miR-146a gene rs2431697 polymorphism was not associated with MS.
  • A significant association was found between the miR-146a gene rs2910164 polymorphism and MS (P = 0.012).

Conclusions:

  • The miR-146a gene rs2910164 polymorphism may contribute to MS susceptibility in the Iranian population.
  • Further research is warranted to elucidate the role of miR-146a in MS pathogenesis.

MicroRNAs

MicroRNA (miRNA) are short, regulatory RNA transcribed from introns (non-coding regions of a gene) or intergenic regions (stretches of DNA present between genes). Several processing steps are required to form biologically active, mature miRNA. The initial transcript, called primary miRNA (pri-mRNA), base-pairs with itself, forming a stem-loop structure. Within the nucleus, an endonuclease enzyme, called Drosha, shortens the stem-loop structure into hairpin-shaped pre-miRNA. After the pre-miRNA...

Single Nucleotide Polymorphisms-SNPs

A single nucleotide polymorphism or SNP is a single nucleotide variation at a specific genomic position in a large population. It is the most prevalent type of sequence variation found in the human genome. Point mutations that occur in more than 1% of the population qualify as SNPs. These are present once every 1000 nucleotides on an average in the human genome. Replacement of a purine with another purine (A/G) or a pyrimidine with another pyrimidine (C/T) is known as a transition. In contrast,...

Genome-wide Association Studies-GWAS

Genome-wide association studies or GWAS are used to identify whether common SNPs are associated with certain diseases. Suppose specific SNPs are more frequently observed in individuals with a particular disease than those without the disease. In that case, those SNPs are said to be associated with the disease. Chi-square analysis is performed to check the probability of the allele likely to be associated with the disease.
GWAS does not require the identification of the target gene involved in...

lncRNA - Long Non-coding RNAs02:39

lncRNA - Long Non-coding RNAs

In humans, more than 80% of the genome gets transcribed. However, only around 2% of the genome codes for proteins. The remaining part produces non-coding RNAs which includes ribosomal RNAs, transfer RNAs, telomerase RNAs, and regulatory RNAs, among other types. A large number of regulatory non-coding RNAs have been classified into two groups depending upon their length – small non-coding RNAs, such as microRNA, which are less than 200 nucleotides in length, and long non-coding RNA...
