High expression of MIR34a is linked to trisomic spontaneous abortions, particularly in cases with advanced maternal age. This finding suggests a potential role for this micro-RNA in pregnancy loss etiology.
Area of Science:
- Reproductive biology
- Epigenetics
- Genetics
Background:
- Spontaneous abortions have complex and varied causes.
- Epigenetic factors, such as micro-ribonucleic acids (miRNAs), may influence pregnancy loss.
- Understanding the molecular mechanisms of miscarriage is crucial for improving reproductive outcomes.
Purpose of the Study:
- To investigate differences in placental miRNA expression between spontaneous abortions with normal and abnormal karyotypes.
- To evaluate specific inhibitor and activator miRNAs in relation to abortion causes and maternal age.
Main Methods:
- A case-control study involving 100 spontaneous abortion samples (disomies, aneuploidies, triploidies).
- Quantitative real-time polymerase chain reaction (qRT-PCR) was used to measure miRNA expression levels.
- Expression of MIR378a-5p, MIR376c, MIR195, MIR34a, and MIR210 was assessed relative to MIR130.
Main Results:
- Placental inhibitor MIR34a expression was significantly higher in trisomic abortion materials (trisomy 16 and 21) compared to disomic controls.
- MIR195 and MIR34a expression levels were elevated in numerically abnormal cases associated with advanced maternal age.
Conclusions:
- High MIR34a expression appears interrelated with trisomic spontaneous abortions.
- Advanced maternal age may exacerbate the effect of MIR34a in trisomic pregnancy losses.
