Summary

Interferon regulatory factor 6 (IRF6) is upregulated in ovarian cancer (OC), correlating with poor prognosis. Knocking down IRF6 inhibits OC cell growth and invasion, suggesting IRF6 as a potential biomarker and therapeutic target for OC.

Background:

  • Ovarian cancer (OC) presents significant diagnostic and therapeutic challenges, particularly serous OC, which lacks effective biomarkers.
  • Identifying novel biomarkers is crucial for improving OC diagnosis and patient care.
  • This study aimed to discover new OC biomarkers and elucidate their pathogenic roles.

Purpose of the Study:

  • To identify novel diagnostic and prognostic biomarkers for ovarian cancer (OC).
  • To investigate the pathogenic role of identified biomarkers in OC development and progression.
  • To explore potential therapeutic targets for OC based on novel biomarker findings.

Main Methods:

  • Utilized differentially expressed genes (DEGs) from "Limma" package and WGCNA modules for intersection analysis in OC.
  • Identified three hub genes (CLDN3, IRF6, PRSS8) via PPI network and validated in multiple public databases (GSE14407, GSE18520, GSE66957, TCGA+GTEx).
  • Confirmed IRF6 expression and prognostic correlation using RT-qPCR, IHC, Kaplan-Meier Plotter, and functional assays (Transwell, scratch wound).

Main Results:

  • Interferon regulatory factor 6 (IRF6) RNA and protein levels were significantly upregulated in serous OC.
  • Elevated IRF6 expression correlated with poorer Overall Survival (OS) and Progression-Free Survival (PFS).
  • IRF6 knockdown suppressed proliferation, migration, and invasion in SKOV3 ovarian cancer cells.

Conclusions:

  • IRF6 is significantly associated with ovarian cancer (OC) development and progression.
  • IRF6 demonstrates potential as a novel biomarker for OC diagnosis and prognosis.
  • IRF6 represents a promising therapeutic target for ovarian cancer patients.
