An ischemic stroke occurs when a cerebral blood vessel becomes obstructed, most often by a thrombus or embolus, interrupting the delivery of oxygen and glucose to brain tissue. Background:

  • Focal cerebral ischemia, or stroke, remains a leading cause of death and disability worldwide.
  • Current treatments for stroke are limited, highlighting the need for novel therapeutic strategies.
  • Inert gases have shown potential in various medical applications, but their neuroprotective effects in stroke are underexplored.

Purpose of the Study:

  • To investigate the neuroprotective effects of krypton-oxygen gas mixture inhalation in a rat model of photoinduced ischemic stroke (PIS).
  • To evaluate the impact of krypton on neurological recovery, brain damage, and key molecular mechanisms post-stroke.
  • To explore the potential of krypton as a therapeutic agent for ischemic stroke.

Main Methods:

  • Male Wistar rats (n=32) were subjected to photoinduced ischemic stroke (PIS).
  • Animals inhaled either a krypton-oxygen (Kr 70%/O2 30%) or a nitrogen-oxygen (N2 70%/O2 30%) mixture for 2 hours.
  • Neurological functions, lesion volume (MRI), cell counts (Iba-1, caspase-3, NeuN), neoangiogenesis (von Willebrand factor), and molecular pathways (AKT, GSK3b, Nrf2, NFkB) were analyzed.

Main Results:

  • Krypton inhalation significantly improved neurological status and reduced lesion volume and cell death (caspase-3 positive cells) in PIS rats.
  • Krypton promoted neoangiogenesis and reduced neuronal loss in the penumbra zone.
  • Krypton inhalation modulated key signaling pathways, increasing phosphorylated AKT, pGSK3b, and Nrf2 expression while decreasing NFkB (p50) expression.

Conclusions:

  • Krypton inhalation demonstrates significant neuroprotective effects in a rat model of focal cerebral ischemia.
  • The observed neuroprotection is associated with reduced apoptosis, enhanced angiogenesis, and modulation of AKT/GSK3b and Nrf2/NFkB signaling pathways.
  • Krypton represents a promising therapeutic candidate for treating ischemic stroke.
