Sulfarotene reverses natural killer (NK) cell-induced stemness in colorectal cancer (CRC) cells, overcoming treatment resistance. This suggests sulfarotene as a potential therapy to enhance NK-cell-mediated immune surveillance against CRC.
Area of Science:
- Oncology
- Immunology
- Cancer Stem Cell Biology
Background:
- Tumor cell stemness drives colorectal cancer (CRC) recurrence, metastasis, and mortality.
- Immune-active cells, like natural killer (NK) cells, can induce tumor cell stemness, leading to treatment resistance.
Purpose of the Study:
- To investigate the effect of NK cell co-culture on CRC cell stemness.
- To evaluate the efficacy of sulfarotene in reversing NK cell-induced stemness in CRC.
- To assess sulfarotene's potential in overcoming chemotherapy resistance in CRC.
Main Methods:
- Co-culture of CRC cell lines with NK cells, with and without sulfarotene.
- Assessment of stemness markers in CRC cells.
- Chemotherapy resistance assays using oxaliplatin and irinotecan.
- In vivo evaluation using a CRC xenograft model.
Main Results:
- NK cell co-culture significantly increased stemness markers in CRC cells.
- CRC cells exhibited enhanced resistance to oxaliplatin and irinotecan post-NK cell co-culture.
- Sulfarotene effectively reversed NK cell-induced stemness in both in vitro and in vivo models.
- Sulfarotene demonstrated efficacy in overcoming chemotherapy resistance.
Conclusions:
- Sulfarotene is a promising agent for reversing NK cell-induced stemness in colorectal cancer.
- Sulfarotene can overcome treatment resistance associated with NK cell activation in CRC.
- This study highlights sulfarotene's potential as an immunotherapeutic strategy to enhance NK-cell-mediated anti-cancer immunity.
