Chimeric antigen receptor (CAR) T-cell therapy shows promise for clear cell renal cell carcinoma (ccRCC). A CD70-targeting CAR T-cell product, CTX130, demonstrated safety and efficacy in a phase I trial, achieving durable responses.
Area of Science:
- Oncology
- Immunotherapy
- Cellular Therapy
Background:
- Clear cell renal cell carcinoma (ccRCC) has limited treatment options.
- Chimeric antigen receptor (CAR) T-cell therapy is a potential novel approach for solid tumors.
Purpose of the Study:
- To evaluate the safety and efficacy of CTX130, an allogeneic CD70-targeting CAR T-cell product, in patients with advanced or refractory ccRCC.
- To explore the potential of CD70-targeted CAR T-cells for treating ccRCC and other CD70+ malignancies.
Main Methods:
- Preclinical studies of CTX130 in RCC xenograft models.
- Phase I, multicenter, first-in-human clinical trial of CTX130 in 16 patients with relapsed/refractory ccRCC.
- Evaluation of a next-generation CAR T construct, CTX131, in preclinical studies.
Main Results:
- CTX130 demonstrated favorable preclinical proliferation, cytotoxicity, and complete regression of RCC xenograft tumors.
- In the phase I trial, no dose-limiting toxicity was observed, and 81.3% of patients achieved disease control.
- One patient achieved a durable complete response lasting 3 years. CTX131 showed improved preclinical expansion and efficacy.
Conclusions:
- Allogeneic CD70-targeted CAR T-cells represent a proof of concept for treating ccRCC and other CD70+ malignancies.
- This study provides the first complete response in a patient with RCC, highlighting the potential of CAR T-cell therapy in solid tumors.
