Allergic reactions related to drugs are hypersensitivity responses driven by the immune system and bear no connection to the drug's therapeutic action. Summary

This study reveals a causal link between eight blood metabolites and allergic conjunctivitis (AC). These findings offer new insights for predicting, preventing, and treating AC by understanding its metabolic underpinnings.

Background:

  • Allergic conjunctivitis (AC) is a prevalent condition with poorly understood metabolic links.
  • Existing research has not fully explored the metabolic basis of AC.

Purpose of the Study:

  • To investigate the causal relationship between blood metabolites and allergic conjunctivitis.
  • To identify specific metabolites associated with AC risk using genetic association data.

Main Methods:

  • Applied two-sample Mendelian Randomization (MR) and Linkage Disequilibrium Score Regression (LDSC).
  • Analyzed data from 20,958 AC cases and 356,319 controls from the metabolomics GWAS server and FinnGen project.
  • Investigated 486 blood metabolites for their association with AC risk.

  • Identified 18 metabolites significantly associated with AC risk using MR, with 16 remaining significant after sensitivity analyses.
