Purpose of the Study:

  • To investigate the potential causal relationship between depression and various hepatobiliary diseases using Mendelian randomization.
  • To explore mediating factors in the causal pathway from depression to non-alcoholic fatty liver disease.

Main Methods:

  • Genome-wide association studies (GWAS) data were used to select genetic variations for depression as instrumental variables.
  • Univariate and multivariate Mendelian randomization (MR) analyses were performed.
  • Sensitivity analyses included Cochran's Q, MR Egger, and Leave-one-out methods.

Main Results:

  • Depression significantly increases the risk of non-alcoholic fatty liver disease (NAFLD), cholelithiasis, chronic pancreatitis, and cholecystitis.
  • Hepatobiliary diseases did not show a causal effect on depression.
  • Waist-hip ratio, hypertension, and daytime naps mediated 35.8% of the causal effect of depression on NAFLD.

Conclusions:

  • Depression is a susceptibility factor for non-alcoholic fatty liver disease.
  • Genetic predisposition to depression causally influences NAFLD risk, with mediation by metabolic and lifestyle factors.
