This study found that YAP, WWTR1, HCP5, SNHG15, and LINC01433 are upregulated in breast cancer (BC). This panel of RNAs shows potential for BC diagnosis and management.
Area of Science:
- Oncology
- Molecular Biology
- Biochemistry
Background:
- Breast cancer (BC) remains a significant public health issue despite declining mortality rates.
- The Hippo pathway, including YAP and WWTR1, is implicated in BC development and progression.
- Long non-coding RNAs (lncRNAs) like SNHG15, HCP5, and LINC01433 are increasingly recognized for their roles in cancer.
Purpose of the Study:
- To investigate the relationship between specific lncRNAs (SNHG15, HCP5, LINC01433) and YAP/WWTR1 in the Hippo pathway.
- To determine the impact of these lncRNAs on YAP and WWTR1 protein expression in breast cancer.
- To explore the potential of these molecules as biomarkers for BC diagnosis and treatment.
Main Methods:
- Analysis of 40 breast cancer tissue samples for RNA and protein expression.
- Quantitative real-time PCR and Western blotting were used to assess mRNA and protein levels.
- Statistical analysis (Mann-Whitney U, Student's t-test) and GEO database validation were performed.
Main Results:
- Upregulation of YAP, WWTR1, HCP5, SNHG15, and LINC01433 at both mRNA and protein levels in BC tissues, consistent with GEO data.
- YAP expression correlated significantly with histological grade; WWTR1 correlated with family history and HER-2 status.
- A combined panel of these five RNAs demonstrated potential for BC prediction via ROC curve analysis.
Conclusions:
- The distinct expression patterns of YAP, WWTR1, SNHG15, HCP5, and LINC01433 in BC tissues highlight their oncogenic roles.
- This panel of five RNAs shows promise as a diagnostic and prognostic biomarker for breast cancer.
- Further investigation into these molecules could lead to novel therapeutic strategies for BC management.

Loss of Tumor Suppressor Gene Functions
When the tumor suppressor genes develop mutations or are lost, cells start growing out of control, leading to cancer. However, a single functional copy of the tumor suppressor gene is enough for the cells to maintain their normal functions and cell...
