    Background:

    • Human Leukocyte Antigen (HLA) class II molecules play a crucial role in immune responses.
    • Characterizing HLA class II diversity is essential for transplantation and autoimmune disease research.

    Purpose of the Study:

    • To isolate and analyze HLA class II antigens from consanguineous homozygous typing cells.
    • To investigate charge heterogeneity and beta chain patterns for HLA class II typing.

    Main Methods:

    • Sequential immunoprecipitation using specific monoclonal antibodies (MoAbs).
    • Analysis of isolated class II antigens on 1D-Isoelectric Focusing (IEF) gels.
    • Assessment of charge heterogeneity and beta chain isoelectric points (pI).

    Main Results:

    • Two to three families of class II antigens were isolated based on DR serotype.
    • Charge heterogeneity demonstrated distinct electrophoretic beta chain patterns among different haplotypes.
