Genome-wide Association Studies-GWAS01:11

Genome-wide Association Studies-GWAS

13.4K
Genome-wide association studies or GWAS are used to identify whether common SNPs are associated with certain diseases. Suppose specific SNPs are more frequently observed in individuals with a particular disease than those without the disease. In that case, those SNPs are said to be associated with the disease. Chi-square analysis is performed to check the probability of the allele likely to be associated with the disease.
GWAS does not require the identification of the target gene involved in...

Human Genetics

565
Human genetics provides a profound framework for understanding the interplay between genetic predispositions and human psychology. At the heart of this discipline lies the study of how genes influence physical traits, behaviors, and susceptibility to diseases. Each person carries a unique genetic code that subtly or significantly shapes their psychological and behavioral landscape.
The complex relationship between genetics and psychology is observable through common biological components such...

Autoimmune Disorders01:29

Autoimmune Disorders

421
Autoimmune diseases are a group of disorders in which the body's immune system mistakenly attacks its own cells, tissues, and organs. This results from an overactive immune response against substances and tissues normally present in the body. Let's delve into the concept and mechanism of autoimmune diseases from an immune system point of view, explore different causes and examples of such diseases, and discuss potential solutions.

Causality in Epidemiology01:21

Causality in Epidemiology

400
Causality or causation is a fundamental concept in epidemiology, vital for understanding the relationships between various factors and health outcomes. Despite its importance, there's no single, universally accepted definition of causality within the discipline. Drawing from a systematic review, causality in epidemiology encompasses several definitions, including production, necessary and sufficient, sufficient-component, counterfactual, and probabilistic models. Each has its strengths and...

Incomplete Dominance01:43

Incomplete Dominance

22.5K
Gregor Mendel's work (1822 - 1884) was primarily focused on pea plants. Through his initial experiments, he determined that every gene in a diploid cell has two variants called alleles inherited from each parent. He suggested that amongst these two alleles, one allele is dominant in character and the other recessive. The combination of alleles determines the phenotype of a gene in an organism.

Summary

This study used Mendelian randomization to investigate the causal link between multiple sclerosis (MS) and psoriasis. Genetic evidence suggests MS reduces psoriasis risk, and psoriasis also reduces MS risk.

Area of Science:

  • Genetics
  • Immunology
  • Epidemiology

Background:

  • Observational studies suggest a bidirectional association between multiple sclerosis (MS) and psoriasis.
  • These associations may be influenced by confounding factors, necessitating causal inference methods.

Purpose of the Study:

  • To examine the directionality of causation between MS and psoriasis using Mendelian randomization.
  • To estimate the genetic correlation between MS and psoriasis.

Main Methods:

  • Mendelian randomization analysis was performed using large-scale genome-wide association studies datasets.
  • Causal analysis using summary effect (CAUSE), inverse variance-weighted (IVW), and pleiotropy-robust methods were employed.
  • Linkage disequilibrium score regression was used to estimate genetic correlation.

Main Results:

  • Mendelian randomization indicated that genetic liability to MS reduces psoriasis risk (OR 0.93, p < 0.05).
  • Conversely, genetic liability to psoriasis reduces MS risk (OR 0.71, p < 0.001).
  • Linkage disequilibrium score analysis revealed no significant genetic correlation between MS and psoriasis (rg = -0.071, p = 0.2852).

Conclusions:

  • There is genetic evidence supporting a bidirectional causal relationship between MS and psoriasis.
  • The findings suggest that MS and psoriasis may influence each other's risk through distinct genetic pathways.
