Tumor Immunotherapy01:27

Tumor Immunotherapy

520
Immunotherapy is a treatment that boosts or manipulates the immune system to fight diseases, including cancer. For instance, by stimulating an immune response through vaccinations against viruses that cause cancers, like hepatitis B virus and human papillomavirus, these diseases can be prevented. Nonetheless, some cancer cells can avoid the immune system due to their rapid mutation and division. The immune response to many cancers involves three phases: elimination, equilibrium, and escape.

Area of Science:

  • Oncology
  • Immunotherapy
  • Cellular Therapy

Background:

  • Chimeric antigen receptor T cell (CAR T cell) therapy is a successful adoptive cell therapy for hematological cancers.
  • Its application in solid tumors is hindered by challenges like cell trafficking, survival in hostile tumor microenvironments, and antigen identification.

Purpose of the Study:

  • To review the challenges of CAR T cell therapy in solid tumors.
  • To present optimizations and novel approaches for enhancing CAR T cell efficacy in solid cancer treatment.

Main Methods:

  • Review of current literature on CAR T cell therapy for solid tumors.
  • Analysis of strategies for improving cell localization, survival, and antigen recognition.

Main Results:

  • Solid tumors present unique challenges including tumor microenvironment hostility and antigen heterogeneity.
  • Emerging strategies focus on precise cell localization, microenvironment modulation, and adaptive antigen recognition.

Conclusions:

  • Overcoming current limitations is crucial for unlocking the full therapeutic potential of CAR T cells in solid tumors.
  • Continued research into advanced CAR T cell designs and delivery methods is essential for clinical success.
