We developed GalTag, a novel proximity labeling technology, to track cellular interactions in solid tissues. This method enables in situ tag generation, overcoming limitations of traditional methods and revealing Bacillus Calmette-Guérin invasion dynamics in mouse bladder tissue.
Area of Science:
- Cell Biology
- Biotechnology
- Biochemistry
Background:
- Understanding cellular interactions is crucial for physiology and pathology.
- Existing proximity labeling methods struggle in solid tissues due to tag diffusion limitations.
Purpose of the Study:
- To develop a novel proximity labeling technology for in situ cellular interaction recording in 3D biological solids.
- To overcome the limitations of diffusion-dependent labeling in solid tissues.
Main Methods:
- Developed GalTag technology utilizing galactose oxidase (GAO) for in situ bio-orthogonal aldehyde tag generation.
- Applied GalTag to monitor cellular interaction dynamics and assess engineered cell targeting.
- Utilized GalTag to visualize Bacillus Calmette-Guérin (BCG) invasion in mouse bladder tissue.
Main Results:
- GalTag successfully generated in situ tags within 3D biological solids.
- The technology enabled real-time monitoring of cellular interaction dynamics.
- First-time recording of BCG invasion footprints in living mouse bladder tissue was achieved.
Conclusions:
- GalTag offers a robust solution for studying cellular interactions in challenging solid tissue environments.
- This technology provides valuable insights into biological processes, such as BCG's anti-tumor mechanisms.

Protein Dynamics in Living Cells
Fluorescent recovery after photobleaching (FRAP) is a fluorescent-protein-based detection technique used to quantify protein movement rates within the cell. This method exposes a small portion of the cell to an intense laser beam. The laser beam causes permanent photobleaching of the fluorophore-tagged proteins in the exposed region. As the bleached...
