Molecular Chaperones and Protein Folding03:00

Molecular Chaperones and Protein Folding

17.9K
The native conformation of a protein is formed by interactions between the side chains of its constituent amino acids. When the amino acids cannot form these interactions, the protein cannot fold by itself and needs chaperones. Notably, chaperones do not relay any additional information required for the folding of polypeptides; the native conformation of a protein is determined solely by its amino acid sequence. Chaperones catalyze protein folding without being a part of the folded protein.

Covalently Linked Protein Regulators

6.8K
Proteins can undergo many types of post-translational modifications, often in response to changes in their environment. These modifications play an important role in the function and stability of these proteins. Covalently linked molecules include functional groups, such as methyl, acetyl, and phosphate groups, and also small proteins, such as ubiquitin. There are around 200 different types of covalent regulators that have been identified.
These groups modify specific amino acids in a protein....

Area of Science:

  • Biochemistry
  • Molecular Biology
  • Cellular Biology

Background:

  • Small heat shock proteins (sHSPs) are vital molecular chaperones involved in cellular stress.
  • sHSPs play significant roles in the pathogenesis of various human diseases.
  • Post-translational modifications (PTMs) are known to affect sHSP function.

Purpose of the Study:

  • To explore the potential of small molecules in modulating sHSP activity.
  • To investigate the link between PTMs of sHSPs and their functional regulation.

Main Methods:

  • Characterization of direct effects of PTMs on specific sHSPs.
  • Exploration of small molecule-mediated modulation of sHSP-associated PTMs.

Main Results:

  • Established that PTMs directly impact the function of certain sHSPs.
  • Demonstrated the feasibility of indirectly regulating sHSP activity via small molecules targeting PTMs.

Conclusions:

  • sHSP activity can be modulated by targeting their post-translational modifications.
  • Small molecule-based strategies hold promise for therapeutic interventions in diseases involving sHSPs.
