    Background:

    • Human interleukin-1 (IL-1) is a key cytokine involved in immune responses.
    • Previous purification methods for IL-1 yielded variable results and lower activity.

    Purpose of the Study:

    • To develop a simplified and efficient procedure for purifying biologically active human IL-1.
    • To characterize the purified human IL-1 protein.

    Main Methods:

    • Purification of IL-1 from stimulated human peripheral blood macrophages using ion exchange and affinity chromatography (Procion Red agarose).
    • Assays for biological activity (thymocyte mitogenesis) and pyrogenicity.
    • Analysis of molecular weight, charge heterogeneity (2D electrophoresis/electrofocusing), amino acid composition, and presence of carbohydrates.

    Main Results:

    • A simplified purification procedure yielded highly pure human IL-1 with excellent biological activity (3.2 X 10(8) U/mg).
    • Purified IL-1 showed no molecular weight heterogeneity, unlike previous reports.
    • Isoelectric focusing revealed charged species (pI 6.0-4.9) with a consistent molecular weight of ~17,500.
    • Amino acid analysis indicated abundant acidic residues and minimal cysteine; no glycosylation was detected.

    Conclusions:

    • The developed method provides a reliable source of pure, active human IL-1.
    • The characterization provides insights into the physicochemical properties of human IL-1.
    • This purified IL-1 is suitable for further research into its biological functions and therapeutic potential.
