Area of Science:

  • Microbiology
  • Molecular Biology
  • Drug Discovery

Background:

  • Antibiotic resistance is a major global health threat, driven by plasmid conjugation in bacteria.
  • New strategies are urgently needed to combat the spread of antimicrobial resistance.

Purpose of the Study:

  • To develop and apply a high-throughput screening method to identify compounds that inhibit plasmid conjugation.
  • To discover novel inhibitors of bacterial conjugation for potential therapeutic use.

Main Methods:

  • A library of 3500 compounds was screened using *Escherichia coli* donor and recipient strains.
  • High-throughput screening involved spotting bacterial cultures treated with compounds onto agar plates for visual assessment.
  • Compounds showing an effect on conjugation were retested for confirmation.

Main Results:

  • Six compounds were identified that demonstrated an ability to inhibit or reduce plasmid conjugation.
  • These compounds represent potential leads for developing new antimicrobial resistance strategies.

Conclusions:

  • The developed high-throughput screening method is effective in identifying conjugation inhibitors.
  • The identified compounds warrant further investigation as potential agents to combat antibiotic resistance.
