Taurine deficiency worsens ulcerative colitis (UC) in mice. Supplementing with taurine protects the gut barrier and reduces inflammation by inhibiting the TLR4/NF-κB pathway, suggesting taurine is a promising treatment for colitis.
Area of Science:
- Gastroenterology
- Immunology
- Nutritional Science
Background:
- Taurine (Tau) is a semiessential amino acid with known benefits for intestinal disorders.
- The precise role of taurine in ulcerative colitis (UC) pathogenesis remains largely undefined.
Purpose of the Study:
- To investigate the influence of taurine on the progression of ulcerative colitis (UC).
- To elucidate the mechanisms by which taurine modulates intestinal inflammation and barrier function.
Main Methods:
- Utilized taurine-deficient mouse models (CSAD-/- and TauT-/-) and dextran sulfate sodium (DSS)-induced colitis.
- Examined the effects of taurine on Caco-2 cells stimulated with lipopolysaccharide (LPS).
- Assessed intestinal barrier integrity, inflammatory markers, and the TLR4/NF-κB signaling pathway.
Main Results:
- Taurine deficiency exacerbated DSS-induced colitis, increasing clinical symptoms and pathological damage.
- Taurine supplementation enhanced intestinal barrier function by increasing goblet cells and tight junction protein expression.
- Taurine inhibited the TLR4/NF-κB pathway, reducing pro-inflammatory cytokines (TNF-α, IL-6) and oxidative stress.
Conclusions:
- Taurine plays a crucial role in maintaining intestinal barrier integrity and suppressing intestinal inflammation.
- Taurine demonstrates potential as a therapeutic supplement for treating colitis.
