    Summary

    Thioglycollate-induced macrophages selectively impact tumor cell growth and metastasis. Repeated exposure led to new tumor variants with altered properties and immune responses, suggesting macrophages can induce tumor evolution.

    Background:

    • Macrophages play a complex role in cancer, influencing tumor growth and immune response.
    • Thioglycollate-elicited peritoneal macrophages (T-PM phi) are a model for studying macrophage functions.

    Purpose of the Study:

    • To investigate the selective effects of T-PM phi on 3LL tumor cells.
    • To determine if T-PM phi can induce changes in tumor cell properties over serial transplant generations.

    Main Methods:

    • 3LL tumor cells were co-transplanted with T-PM phi over multiple generations (6-15) in C57BL/6 mice.
    • Tumor growth, metastasis, and immune responses were assessed in vivo and in vitro.
    • Characterization of resulting tumor variants (3LLR6, 3LLR15) for morphological, tumorigenic, and antigenic properties.
