    Background:

    • Oocyte maturation is a critical process in sexual reproduction.
    • Understanding the molecular mechanisms that drive oocyte maturation is essential.

    Purpose of the Study:

    • To investigate the presence and activity of maturation-promoting factor (MPF) during mouse oocyte maturation.
    • To determine if MPF activity can induce germinal vesicle breakdown in a heterologous oocyte system.

    Main Methods:

    • Cytoplasmic extracts from mature and immature mouse oocytes were prepared.
    • These extracts were injected into immature Xenopus laevis oocytes.
    • The effect of extracts on germinal vesicle breakdown (GVBD) was observed.
    • The role of protein synthesis was assessed using cycloheximide.

    Main Results:

    • Injection of mature mouse oocyte extracts induced rapid germinal vesicle breakdown in Xenopus oocytes.
    • This induction was independent of protein synthesis, as cycloheximide did not inhibit the process.
    • Extracts from immature mouse oocytes did not induce germinal vesicle breakdown.

    Conclusions:

    • Maturation-promoting factor activity is present in the cytoplasm of meiotically mature mouse oocytes.
    • This activity is sufficient to induce germinal vesicle breakdown in Xenopus oocytes.
    • MPF activity emerges during the progression of oocyte maturation in mice.
