    Background:

    • Natural killer (NK) cells are crucial for innate immunity.
    • Human colostrum, the first milk produced after birth, contains immune cells and factors that protect newborns.

    Purpose of the Study:

    • To investigate the presence and characteristics of NK cells in human colostrum.
    • To determine the functional activity of colostral NK cells and their interaction with colostrum components.

    Main Methods:

    • Monoclonal antibody HNK-1 (Leu-7) was used to identify NK cells in colostrum.
    • Fluorescence-activated cell sorting (FACS) isolated HNK-1-positive cells.
    • Electron microscopy examined cell morphology.
    • Cytotoxicity assays assessed NK cell activity against K562 target cells.

    Main Results:

    • Approximately 0.5% of colostral cells were identified as NK cells (HNK-1 positive).
    • Two distinct morphological types of colostral NK cells were observed.
    • Colostral NK cells exhibited low cytotoxic activity.
    • Incubation with cell-free colostrum inhibited the cytotoxic activity of peripheral blood NK cells in a dose-dependent manner, causing morphological changes.

    Conclusions:

    • Human colostrum contains NK cells with reduced cytotoxic function.
    • Colostrum possesses humoral factors that can modulate NK cell morphology and suppress their activity.
