Defense Against Bacterial Pathogens01:31

Defense Against Bacterial Pathogens

1.4K
The human immune system is a complex network of cells, tissues, and organs that work together to defend the body against bacterial infections. It consists of various immune cells, each playing a specific role in the defense mechanism.
Phagocytes
Phagocytes are the frontline soldiers of the immune system. They include neutrophils and macrophages. Neutrophils are the most abundant type of white blood cell and are quickly mobilized to the site of infection. Macrophages are larger cells that patrol...

Surface Membrane Barriers01:18

Surface Membrane Barriers

1.1K
The skin and mucous membranes serve as the primary line of defense against pathogens by providing both physical and chemical protection. These barriers are essential in preventing the entry and establishment of microbes, thereby maintaining the integrity of the host.
The outer layer of the skin, the epidermis, is a robust barrier comprising layers of closely packed keratinized cells. This dense arrangement prevents microbes from penetrating the body. The periodic shedding of epidermal cells...

Combined Effects of Drugs: Synergism

3.8K
Synergism is a useful mechanism where combining two or more drugs is more effective than each constituent used alone. Such combinations are also called supra-additive interactions. The drugs collectively enhance the final therapeutic effect by acting on different targets. Another advantage is that the low dose of each constituent drug is sufficient to achieve the desired effect. This helps reduce the duration of therapy and lower the adverse effects of these drugs.
Such synergistic combinations...

Summary

Rhodomyrtone shows potent activity against methicillin-resistant Staphylococcus aureus (MRSA), including strains resistant to other antibiotics. This plant-derived compound may offer a new treatment option for challenging MRSA infections.

Area of Science:

  • Microbiology
  • Pharmacology
  • Natural Products Chemistry

Background:

  • Rhodomyrtone is a novel antibiotic derived from the plant Rhodomyrtus tomentosa.
  • It presents a potential alternative in combating antibiotic-resistant bacteria.

Purpose of the Study:

  • To assess rhodomyrtone's efficacy against various methicillin-resistant Staphylococcus aureus (MRSA) strains.
  • To evaluate its activity against MRSA isolates with existing resistance to vancomycin, daptomycin, linezolid, and ceftaroline.

Main Methods:

  • Broth microdilution testing was employed to determine Minimum Inhibitory Concentrations (MICs) and Minimum Bactericidal Concentrations (MBCs).
  • Rhodomyrtone's activity was compared against fosfomycin, vancomycin, daptomycin, linezolid, and ceftaroline.

Main Results:

  • Rhodomyrtone achieved an MIC90 of 1 mg/L against 110 MRSA strains, including those resistant to daptomycin, vancomycin, and linezolid.
  • The MBC90 for rhodomyrtone was 4 mg/L across all tested MRSA groups, not exceeding 8 mg/L for any S. aureus group.

Conclusions:

  • Rhodomyrtone exhibits significant antimicrobial activity against MRSA, including multidrug-resistant strains.
  • Its effectiveness against isolates with reduced susceptibility to established anti-MRSA drugs suggests potential clinical applications for treating difficult MRSA infections.
