RNA Editing02:23

RNA Editing

9.0K
RNA editing is a post-transcriptional modification where a precursor mRNA (pre-mRNA) nucleotide sequence is changed by base insertion, deletion, or modification. The extent of RNA editing varies from a few hundred bases, in mitochondrial DNA of trypanosomes, to a just single base, in nuclear genes of mammals. Even a single base change in the pre-mRNA can convert a codon for one amino acid into the codon for another amino acid or a stop codon. This type of re-coding can significantly affect the...

Genetic Screens02:46

Genetic Screens

4.9K
Genetic screens are tools used to identify genes and mutations responsible for phenotypes of interest. Genetic screens help identify individuals or a group of people at risk of developing  genetic diseases and help them with early intervention, targeted therapy, and reproductive options.
Forward genetic screens
Forward or “classical” genetic screens involve creating random mutations in an organism’s DNA using radiation, mutagens, or insertion of additional bases, which...

Area of Science:

  • Plant Biotechnology
  • Molecular Biology
  • Crop Science

Background:

  • Current methods for screening transgenic soybeans are time-consuming and require specialized equipment.
  • Visual markers are needed for efficient early screening of genetically modified soybean plants.

Purpose of the Study:

  • To evaluate the RUBY reporter system for visual screening of transgenic soybean events.
  • To assess the utility of RUBY for identifying genome-edited, transgene-free soybean seedlings.

Main Methods:

  • The RUBY reporter system, encoding betalain biosynthetic enzymes, was introduced into soybean plants.
  • Visual assessment of purple pigment accumulation was used to confirm transformation.
  • Genetic stability of the transgene was analyzed across generations using cotyledons.

Main Results:

  • The RUBY reporter system successfully enabled visual confirmation of transgenic soybean events.
  • Visible purple pigment accumulation accurately indicated successful transformation.
  • Transgene-free seedlings were easily distinguished by their green color, facilitating selection.

Conclusions:

  • The RUBY reporter system provides an efficient and convenient visual tool for soybean transformation and genome editing.
  • This system accelerates the selection of desirable soybean lines, including transgene-free mutants.
