This study presents an improved RNA extraction method for cotton, effectively removing phenolic compounds using Polyvinyl pyrrolidone polymer (PVPP) and Lithium chloride (LiCl) precipitation. The protocol yields high-quality RNA suitable for analyzing mitochondrial genes in cytoplasmic male sterility (CMS) cotton lines.
