Background:

  • Common wild rice (Oryza rufipogon) is a valuable genetic resource.
  • A stable wild rice inbred line, Huaye3, was previously identified but its drought tolerance was unknown.

Purpose of the Study:

  • To evaluate the drought tolerance of the Huaye3 wild rice line.
  • To compare germination and seedling growth of Huaye3 with Dawennuo under polyethylene glycol (PEG6000)-induced drought stress.

Main Methods:

  • Germination and seedling growth assays under five PEG6000 concentrations (0-20%).
  • Cytological observations of root tips.
  • Whole-genome re-sequencing analysis.

Main Results:

  • Huaye3 exhibited significantly higher germination rates (52.50%) than Dawennuo (25.83%) under 20% PEG6000.
  • Huaye3 showed fewer cellular abnormalities in root tips under drought stress.
  • Re-sequencing identified 39 drought stress-related genes with variations in the CDS region in Huaye3.

Conclusions:

  • Huaye3 possesses strong drought stress tolerance.
  • This study provides a basis for selecting drought-resistant rice germplasm.
